It's a 64 gig card.
Then if you want CHDK to start automatically, you need to format the card FAT32, which you can do with a computer, but not in-camera. On Windows, you may need to use command line or a 3rd party utility.
So, must I start in Auto mode, or will program work?
And, do I still start it using the picture review button?

I'm not clear what you are asking here. You should be able to use Auto or P mode. On some cameras, CHDK raw is not available in Auto, but I don't see any mention of this for Elph 110. None of this is related to CHDK starting automatically ("autoboot") or not.
If you are booting CHDK manually with "Firm update", you must start with the playback button. If autoboot is configured then you should be able to use either, but you will probably have to hold down the power button a bit longer to start in shooting mode.
On Elph 110 the playback button is the alt button, so when CHDK is running, you click that to access CHDK menus, and must hold it longer to get the original function.
