Hello,
I am a researcher working on fluorescence imaging using a Point and Shoot camera. The specifications for the camera are
1. 12 bit RAW images
2. 20 mm working distance
3. Remote shooting and RAW image download over USB

After some research, CHDK seemed to be the solution for remote shooting through the PTP Extension.
However I am confused about which models can give 12 bit RAW images. Most of the P&S cameras have a minimum focus distance of 3cm, so can I still get images from a distance of 20 mm or do I have to get a macro lens.

Any help/suggestions would be appreciated. Thank you.

My SX50 has 12bit RAW the starts with 0mm focus distance.
The IXUS160 has 12bit DNG RAW and starts with 10mm focus distance.

Only very old powershots (roughly digic III and earlier) used 10 bit raw, any vaguely modern camera will be 12 bits (or 14 in the case of G1x)
Don't forget what the H stands for.

I have selected SX530 for its good macro mode and availability. The camera will be enclosed in a box to capture the fluorescence. I am considering using PTP Extension to control the camera from Windows.
Are all the functions accessible over USB and can the RAW images be transferred over USB to PC for analysis using script?
Is the full resolution of the camera supported by the PTP Extension?

I'm not sure exactly what you mean by "all functions" (canon firmware features? CHDK features?), but in general you can control pretty much everything. There are some things which can be inconvenient to configure if the camera is totally inaccessible.

CHDK raw files be downloaded, or captured directly to your PC using the remote shoot functionality. Unlike most ports, the sx530 port does not currently support remote shoot for jpegs, so they will be saved to the card no matter what.

Full resolution of? You can shoot and download images of any supported resolution. The PTP live view has resolution equivalent to what shows on the camera LCD.

It will be like flashing the micro controller with a code which will take a set of RAW images when started and transfer it to PC and then shut down. The focus and exposure settings will be kept constant at optimal settings.

Can the current port of SX530 be used for RAW file transfer to PC with or without saving on SD card?

You can take raw images and transfer to PC and shutdown use CHDK.
Note if the camera is completely inaccessible, you will need some way to power it on, either:
1) Arrange some way to physically press the power or play buttons
2) Physically replace the power with some external control
3) Arrange for the power button to be permanently held down, and control the power supply externally. This method works on most cameras, but I'm not aware of it being tested on sx530.

Setting fixed exposure and focus distance should be no problem.

Yes, either one. On this port, jpeg will always be saved to the card.
