to avoid vignetting i would suggest using constant av value.
in general, the more settings are fixed, the better (stable) the result.
but, if you want to shoot night&day timelapses then that's whole different story.
lcd turned off - that's a must.
external power supply - also. especially if you're planning to do some long timelapses.
resolution - for web publishing 640 is enough i think, but you could shoot in 1280 or 1600 as well, and then crop/resize images.
always use batch processing for your sequences. i use irfanView + virtualdub combo.
delay between shots - depends on what you're shooting.
fps - 24 or 30. i found videos with 60 fps play choppy on online services.
experiment - make some timelapses, learn, find what suits you the best.

a lot of this has been said before, search :)

I agree with 'barret' that "lcd turned off" is important for timelapse.
Unfortunately, it appears that that is not possible with the S5IS brcause set_backlight(d) is non-functional.
Works on my S2IS but not on my S5IS.
Bummer!

Assign "Sleep" to the "Shortcut" (print) button from the Canon menu, then do a simple

press "print"
sleep 100
release "print"

sequence in your script after each shot, this will disable the LCD and also the sensor, this gives max. power saving.
But there can be side effects with this method, e.g. loosing focus; MD (motion detection) will not work while the display is off.

...but

btw - set_backlight() just disables the LCD backlight, the LCD itself, the electronics and the sensor will still be active; the method above results in a longer battery lifetime...

BTW, I posted this elsewhere, but heres my first timelapse besides my failed 15sec cloud timelapse.

NY-FL coming soon... its a [admin: avoid swearing please] to process 10400 pictures properly... just one thing after another goes wrong. First the tilt sensor data screwed up a whole bunch of pictures making the resolutions not consistent when batch converting with Irfanview. Then some other [admin: avoid swearing please] went wrong. But now I got all the pictures ready, just gotta find a good tune. Unfortunately my comp cant preview the timelapse (~20-30) at the correct frame rate, so I can't really see how well the music works.

PS>lol we got some sensitive definition of swearing here. I used a word with a C that also means POO. and the other ones only a female dog
